NCBI Description

This gene is a member of the aggrecan/versican proteoglycan family. The encoded protein is an integral part of the extracellular matrix in cartilagenous tissue and it withstands compression in cartilage. Mutations in this gene may be involved in skeletal dysplasia and spinal degeneration. Multiple alternatively spliced transcript variants that encode different protein isoforms have been observed in this gene. [provided by RefSeq]

Uniprot Description

ACAN iso3: This proteoglycan is a major component of extracellular matrix of cartilagenous tissues. A major function of this protein is to resist compression in cartilage. It binds avidly to hyaluronic acid via an N-terminal globular region. Defects in ACAN are the cause of spondyloepiphyseal dysplasia type Kimberley (SEDK). Spondyloepiphyseal dysplasias are a heterogeneous group of congenital chondrodysplasias that specifically affect epiphyses and vertebrae. The autosomal dominant SEDK is associated with premature degenerative arthropathy. Defects in ACAN are the cause of spondyloepimetaphyseal dysplasia aggrecan type (SEMD-ACAN). A bone disease characterized by severe short stature, macrocephaly, severe midface hypoplasia, short neck, barrel chest and brachydactyly. The radiological findings comprise long bones with generalized irregular epiphyses with widened metaphyses, especially at the knees, platyspondyly, and multiple cervical-vertebral clefts. Defects in ACAN are the cause of osteochondritis dissecans short stature and early-onset osteoarthritis (OD). It is a type of osteochondritis defined as a separation of cartilage and subchondral bone from the surrounding tissue, primarily affecting the knee, ankle and elbow joints. It is clinically characterized by multiple osteochondritic lesions in knees and/or hips and/or elbows, disproportionate short stature and early-onset osteoarthritis. Belongs to the aggrecan/versican proteoglycan family. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The ACAN acan (Catalog #AAA604127) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Aggrecan ARGxx (Proteoglycan, PG, AGC 1, Aggrecan 1, Antigen identified by monoclonal A0122, Cartilage specific proteoglycan core protein, Chondroitin sulfate proteoglycan core protein 1, CSPCP, CSPG1, CSPGCP, Large aggregating proteoglycan, MSK16, SEDK) reacts with Bovine, Canine, Equine, Feline, Guinea Pig, Human, Porcine, Rabbit, Rat, Sheep and may cross-react with other species as described in the data sheet. AAA Biotech's Aggrecan ARGxx can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB), Immunohistochemistry (IHC). Suitable for use in ELISA, Western Blot and Immunohistochemistry. Dilution: Western Blot: 1:50 Detects a variety of epitopes between 50 and 250kD. Immunohistochemistry: Formalin or paraformaldehyde- fixed paraffin embedded sections or alcohol-fixed frozen or unfixed snap-frozen sections. Notes:Samples must be deglycosylated using 0.01 Units Chondroitinase ABC, 0.01 Units Keratanase and 0.0001 Units Keratanase II per 10ug S-GAG of non-deglycosylated aggrecan for optimal epitope recognition. See Little et al. and Caterson et al. Diluted Technomouse culture supernatant. Researchers should empirically determine the suitability of the ACAN acan for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Aggrecan ARGxx, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
